Understanding Type 1 Diabetes through Imaging Mass Cytometry

Type 1 diabetes (T1D) is a complex autoimmune disease characterized by the destruction of insulin-producing beta cells in the pancreas. Researchers are working to gain a better understanding of T1D development and progression, with a focus on identifying specific beta cell subsets that may be more sensitive to destructive signals or responsive to regenerative cues.

In the Bodenmiller lab, we are using imaging mass cytometry to analyze pancreas sections of individuals with or at risk for type 1 diabetes. This cutting-edge technology enables us to multiplex and deeply phenotype beta cells and infiltrating immune cells, as well as map their interactions. By combining high-throughput imaging with advanced data analysis tools, we aim to uncover the molecular mechanisms underlying T1D development.

One of our key approaches is to study how beta cell subpopulations respond to external stimuli relevant to T1D using primary human islet culture and imaging mass cytometry. This allows us to explore the heterogeneity of beta cells and their interactions with infiltrating immune cells in a more comprehensive manner.

Our research has sparked an interest in both T1D research and image analysis of single cells, two elements that remain at the center of our current projects. During my thesis, I used confocal microscopy and lineage tracing in complex transgenic mouse models and primary human pancreatic islets to study beta cell regeneration.

In collaboration with Pedro Herrera at the University of Geneva, we are also exploring the potential for regenerative therapies, such as the transdifferentiation of alpha cells into insulin-producing beta cells. By leveraging advanced imaging technologies and experimental biology techniques, we hope to uncover new insights into T1D development and progression, ultimately leading to more effective treatments for this complex disease.

In conclusion, our research aims to gain a better understanding of T1D development in the pancreas through highly multiplexed imaging, data analysis, and experimental biology. By combining these approaches, we hope to uncover the underlying mechanisms of T1D and identify new therapeutic targets for the treatment of this devastating disease.
